Functional Annotation of the Druggable Proteome Through Covalent Proteogenomics
Most disease-causing proteins remain inaccessible to chemical probes or molecules that can serve as leads for future drugs. We take an interdisciplinary approach that unites proteomics, genomics, and chemical biology to address this gap and to uncover functional, disease-linked, and druggable pockets of human proteins, including those unique to cancers. Our proposal will yield new technology to rapidly connect proteins to chemical probes and a general strategy to bridge the current divide between genetic diagnosis and production of tailored therapies.
